本文观察了地塞米松(Dexa)及全反式维甲酸(Ra)两药对人类多发性骨髓瘤(MM)OPM2的体外作用。所有参数表明两药有抑制克隆生长作用,抑制程度与药物剂量有浓度依赖关系。两药在高浓度有很强的协同抑制作用,低浓度除外。外源性IL6能完全逆转Dexa致生长抑制但对Ra无效。RTPCR测得Dexa能使IL6R密度增高,IL6RmRNA表达降低,而测得Ra则相反。如两药联合测得IL6RmRNA表达更明显降低。但均对受体亲合性有关的gP130mRNA表达无作用。两药均可清除培养上清液中瘤细胞分泌的IL6。因此本研究证明,通过两药协同作用调节骨髓瘤自身/旁分泌IL6,可有力抑制瘤细胞的增殖。
In this paper, the in vitro effects of dexamethasone (Dexa) and all-trans retinoic acid (Ra) on OPM-2 in human multiple myeloma (MM) were observed. All parameters indicate that the two drugs inhibit the growth of clones, and the degree of inhibition has a concentration-dependent drug dose. Two drugs in high concentrations have a strong synergistic inhibitory effect, except for low concentrations. Exogenous IL 6 can completely reverse the growth inhibition caused by Dexa but not effective on Ra. RT-PCR measured Dexa IL 6R density can be increased, IL 6R mRNA expression decreased, while the measured Ra is the opposite. As measured by the two drugs IL 6R mRNA expression was significantly reduced. But all had no effect on the receptor affinity of gP130 mRNA expression. Two drugs can clear the culture supernatant of tumor cells secreted IL 6. Therefore, this study shows that synergy through the two drugs regulate myeloma itself / paracrine IL 6, can effectively inhibit tumor cell proliferation.
