[目的]研究osteoglycin表达与肿瘤转移的关系。[方法]构建真核表达载体pIRE-Spuro3 osteoglycin(+),将其转染至小鼠肝癌细胞株Hca-F,评价osteoglycin表达对Hca-F细胞转移生物学行为的影响。[结果]转染pIRESpuro3 osteoglycin(+)的Hca-F细胞迁移能力和侵袭能力显著下降;体内试验显示,其外周淋巴结转移率明显降低;然而细胞增殖能力及粘连能力未见明显变化。[结论]高表达osteoglycin降低Hca-F细胞淋巴结转移能力。
[Objective] To study the relationship between osteoglycin expression and tumor metastasis. [Method] The eukaryotic expression vector pIRE-Spuro3 osteoglycin (+) was constructed and transfected into Hca-F mouse hepatoma cell line to evaluate the effect of osteoglycin expression on the biological behavior of Hca-F cells. [Results] The migration and invasion ability of Hca-F cells transfected with pIRESpuro3 osteoglycin (+) significantly decreased. In vivo experiments showed that the rate of peripheral lymph node metastasis was significantly decreased. However, no significant changes were observed in cell proliferation and adhesion ability. [Conclusion] Overexpression of osteoglycin reduces the lymph node metastasis of Hca-F cells.
