目的核因子-κB诱导激酶(nuclear factor-κB inducing kinase,NIK)在多种肿瘤细胞中高表达,但其在喉癌中的表达情况目前尚不清楚。为此,本研究探讨NIK蛋白在喉癌组织中的表达及其临床意义。方法收集河北医科大学第三医院2000-01-01-2010-09-30接受手术治疗的63例喉癌患者的术中切除标本,所有患者均经术后病理确诊。采用免疫组织化学方法检测NIK蛋白在喉癌及癌旁正常组织中的表达水平,探讨NIK表达水平与患者的预后和临床病理特征的相关性。结果 NIK在喉癌组织中的阳性表达率为60.3%(38/63),显著高于癌旁正常组织的9.5%(6/63),χ2=35.76,P<0.001。NIK的表达与患者的性别(χ2=0.02,P=0.881)、年龄(χ2=0.027,P=0.867)、分化程度(χ2=0.14,P=0.932)和肿瘤部位(χ2=2.36,P=0.127)无关,而与患者的临床分期(χ2=13.7,P=0.003)和有无淋巴结转移(χ2=20.2,P<0.001)显著相关。NIK阳性表达的喉癌患者5年生存率为28.6%,显著低于阴性表达患者的80.9%,χ2=4.07,P=0.043。多因素Cox比例风险回归模型分析进一步显示,NIK蛋白表达水平为喉癌患者预后的独立影响因素,P=0.022。结论 NIK在喉癌组织中的表达明显升高,可能是影响患者预后的重要因素。
ObjectiveNuclear factor-κB inducing kinase (NIK) is highly expressed in a variety of tumor cells, but its expression in laryngeal carcinoma is not yet clear. To this end, the study to explore the expression of NIK protein in laryngeal cancer and its clinical significance. Methods Totally 63 cases of laryngeal carcinoma underwent surgical resection in the Third Hospital of Hebei Medical University from January 2000 to January 2010. The patients were all undergone pathological examination. Immunohistochemistry was used to detect the expression of NIK protein in laryngeal carcinoma and adjacent normal tissues, and the correlation between NIK expression level and prognosis and clinicopathological features were analyzed. Results The positive rate of NIK expression in laryngeal carcinoma was 60.3% (38/63), which was significantly higher than that in adjacent normal tissues (9.5%, χ2 = 35.76, P <0.001). The expression of NIK was positively correlated with the gender (χ2 = 0.02, P = 0.881), age (χ2 = 0.027, P = 0.867) ), But not with clinical stage (χ2 = 13.7, P = 0.003) and lymph node metastasis (χ2 = 20.2, P <0.001). The 5-year survival rate of NIK-positive laryngeal cancer patients was 28.6%, significantly lower than that of negative expression patients (80.9%, χ2 = 4.07, P = 0.043). Multivariate Cox proportional hazards regression model analysis further showed that NIK protein expression was an independent prognostic factor for laryngeal cancer, P = 0.022. Conclusion The expression of NIK in laryngeal carcinoma was significantly increased, which may be an important factor affecting the prognosis of patients.
