目的 :为了较深入的阐述配位金属离子手性添加剂拆分药物对映体的机理。方法 :本文运用已建立的配位体交换液相色谱 (CLEC)方法 ,以新型配位添加剂 (R ,R) -酒石酸 -单 -n -辛胺 (TAMOA)为研究对象 ,将有关文献进行了总结和综述 ,对其拆分机理及影响因素进行了讨论。结果 :运用这种方法进行手性拆分 ,其机理为 :首先拆分样品与手性添加剂以共价键形成五 (六 )元比较稳定的环状结构 ,同时手性添加剂 (SE) ,金属离子 (Me) ,外消旋分子 (SA)之间形成多种平衡形式的中间配合物 ;然后 ,根据不同配合物在固定相上的保留时间的不同进行拆分。结论 :配位金属离子手性添加剂CLEC方法拆分条件简单 ,成本低 ,可在手性药物分析及手性单一体半制备中有广泛的应用前景
OBJECTIVE: To elucidate the mechanism of resolution of drug enantiomers by coordination metal ion chiral additives. Methods: In this paper, the established ligand exchange liquid chromatography (CLEC) method, the new coordination additive (R, R) - tartaric acid - mono-n-octylamine (TAMOA) Summarize and summarize, discuss its split mechanism and influencing factors.
