目的:探讨高盐环境对小鼠脑缺血后IP-10(Interferon induced protein 10,IP-10)表达的影响。方法:将C57BL/6雄性小鼠分为高盐饮食(4%Na Cl鼠粮和1%Na Cl水)组和正常饮食组,建立永久性大脑中动脉结扎(permanent middle cerebral artery ligation,p MCAL)小鼠模型;激光共聚焦显微镜检测脑缺血组织IP-10表达情况;RT-PCR(Reverse transcription PCR)检测脑缺血组织中IP-10表达情况;ELISA方法检测内皮细胞和神经胶质细胞氧糖剥夺(oxygen-glucose deprivation,OGD)6h后上清中IP-10表达。结果:免疫荧光观察发现高盐饮食组小鼠脑缺血组织中IP-10阳性细胞表达增加;与正常饮食组相比,高盐饮食组缺血侧脑组织中IP-10表达显著增加,有统计学意义(P1d<0.05,P2d<0.001);与内皮细胞相比,神经胶质细胞OGD后,上清中IP-10表达显著增多(P<0.001)。结论:高盐环境促进脑缺血后IP-10表达;脑缺血损伤过程中,脑内的IP-10主要是由胶质细胞释放的。
Objective: To investigate the effect of high salt environment on the expression of IP-10 in mice after cerebral ischemia. Methods: C57BL / 6 male mice were divided into high salt diet (4% NaCl diet and 1% NaCl water group) and normal diet group. Permanent middle cerebral artery ligation (p MCAL ) Mouse model was established. The expression of IP-10 in cerebral ischemic tissue was detected by laser scanning confocal microscopy. The expression of IP-10 in cerebral ischemic tissue was detected by RT-PCR (Reverse transcription PCR). The expression of IP- IP-10 expression in supernatants after 6h of oxygen-glucose deprivation (OGD). Results: Immunofluorescence showed that the expression of IP-10 positive cells increased in the hippocampus of mice in high-salt diet group. Compared with the normal diet group, the expression of IP-10 in the ischemic hippocampus increased significantly (P <0.001). Compared with endothelial cells, the expression of IP-10 in supernatants was significantly increased after glial cells were treated with OGD (P <0.001). Conclusion: High-salt environment can promote the expression of IP-10 after cerebral ischemia. During the process of cerebral ischemia, IP-10 in the brain is mainly released by glial cells.
