Background:The aims of this study are to induce fatty liver in zebrafish larvae by overfeeding approach and to examine the effects of caffeine on hepatic lipid accumulation in overfeeding zebrafish larva.Methods:100 zebrafish larvae from a single cluster at 5dpf were allocated to a series of different amount of feeding groups:20mg/d,30mg/d,60mg/d,80mg/d,120mg/d and 180mg/d and fed for 10,15,and 20 days.By observing the morphological and pathological changes in zebrafish,the growth and the hepatic steatosis in different quantity and duration of feeding group were evaluated.For caffeine treatment,larvae were overfed with 1 %,2.5 %,5 %and 8 %caffeine for 20 days,the effect and mechanism of caffeine on hepatic steatosis in zebrafish were explored via evaluating histological changes in liver and examinating the body weight,lipid contents,and associated genes expression with qRT-PCR.
