拜氏梭菌戊糖代谢工程

  丁醇是重要的大宗化学品,可通过产溶剂梭菌发酵生产.拜氏梭菌对秸秆类木质纤维素水解液的耐受性较好,但对水解液中己糖和戊糖的共发酵能力有待提升.我们改进了原有TargeTron基因失活技术,可方便、有效的连续中断拜氏梭菌基因.对拜氏梭菌NCIMB 8052有机酸和戊糖代谢途进行组合改造,获得的工程菌LJL028几乎完全消耗仿真料液中的葡萄糖和阿拉伯糖,木糖利用率也达到91%,可产丁醇12.46 g/L,总溶剂19.14 g/L,分别比野生型提高46%和37%.结果表明工程菌LJL028是一株有潜力的纤维素丁醇生产菌.
