Hepatocellular carcinoma (HCC) mostly develops in the liver under chronic inflammation.Oxygen radicals produced at the site of inflammation attack membrane bound and circulating lipids forming a number of intermediate metabolites including lipid hydroperoxides.Lipid hydroperoxides can inroporate into the membranes initiating further lipid peroxidation.The role of these compounds in HCC has not been investigated so far.We adressed this issue using linoleic acid hydroperoxides (LOOH) and applying them to the cultured human hepatocarcinoma cells.The addition of LOOH to the medium human hepatocarcinoma cell line (HCC-1.2, HCC-1.3, HUH7, SNU398) caused dosedependent cell loss and enhanced LDH-release.Under subtoxic conditions LOOH induced intracellular hydrogen peroxide production and a decrease of cellular GSH content.Moreover, the cells were pushed into the S-and G2/Mphase by LOOH while unoxidized linoleic acid was not active.
