N-烯基-α,β-不饱和硝酮与异氰酸酯合成二氮杂环壬烷

  九元氮杂环化合物不仅出现在天然与非天然的化合物中,而且也用作合成多种生物碱的有用中间体,而且九元杂环的合成一直也是有机合成领域的一个难点。本课题组一直从事硝酮的合成及其在合成氮杂环骨架中的应用。我们首次以N-烯基-α,β-不饱和硝酮与异氰酸酯为试剂,通过[3+2]环加成反应形成五元氮杂环中间体,进而发生3,3-重排反应,高立体选择性的一锅法构建二氮杂环壬烷骨架。反应底物范围广,官能团的兼容性好,产物的收率较高。进一步研究发现,异氰酸酯上的取代基可以控制反应的非对映选择性。通过手性辅基的策略可以容易地获得光学纯的二氮杂环壬烷。二氮杂环壬烷在不同的反应条件下可以转化成骨架多变的杂环化合物,为此类化合物的进一步研究和应用奠定基础。
