Highly reducing iterative polyketide synthases(HRPKS) are multifunctional,complex enzymes.Hpm8 is a HRPKS responsible for synthesizing the reduced hexaketide portion of dehydrozearalenol(DHZ),an advanced intermediate in the biosynthesis of hypothemycin.We systematically investigated the putative intermediates involved in the formation of the highly reduced hexaketide portion by incorporation of 13C-labeled N-acetylcysteamine(SNAC) thioester intermediates into DHZ using purified Hpm8 and Hpm3.Our results showed that Hpm8 could accept putative intermediates,and assemble them into DHZ through cooperation with Hpm3.In addition,feeding Hpm8 and Hpm3 with structurally modified intermediates could produce DHZ analogues.
