  As the prolactin receptor (PRLR) signaling was proved to be involved in breast and prostate tumorigenesis, there is a real need to develop therapeutic strategies to block its activation.The active complex between prolactin (PRL) and its receptor is a heterotrimer involving one hormone molecule and two receptor moieties.The ligand-receptor interacting interfaces are called binding sites 1 and 2.In order to develop pure PRLR antagonists, we first started with the strategy used for the homologous growth hormone receptor consisting in mutating the Glycine lying at the bottom of the hormone site 2 cavity by an Arginine.
