The huge majority of currently used drugs is based on "organic" compounds not containing metal atoms.However,metal complexes offer interesting opportunities for the development of novel drugs due to their unique properties concerning reactivity,geometry or kinetics and the continuing success story of the platinum antitumor drugs (e.g.cisplatin or carboplatin) demonstrates that metal agents can be powerful therapeutics.Current research in the field of bioinorganic and bioorganometallic medicinal chemistry focuses on the design and development of novel "metallodrugs",which differ substantially from the established platinum agents and are also supposed to overcome the major disadvantages of platinum tumor therapy concerning resistance development and side effects.In this presentation we will present two prototypic examples for new non-platinum antitumor drugs,which showed promising pharmacological properties in early preclinical studies:a hexacarbonyldicobalt complex with a aspirin-derived ligand,which inhibits the aspirin target enzymes (cyclooxygenases) by an altered molecular interaction and a gold-phosphine complex containing a naphthalimide ligand,which inhibits thioredoxin-reductase,induces apoptosis and inhibits angiogenesis.
