目的:通过测定蒙古口蘑多糖(Mongolia Tricholoma polysaccharide,MOP)的抗氧化活性,观察其在低密度脂蛋白(Low density lipoprotein,LDL)被氧化修饰过程中的影响及其在抗内皮细胞凋亡的作用,探讨其可能的保护血管、抗AS的作用机制.方法:将蒙古口蘑子实体用无水乙醇进行脱脂处理后90℃加热6h进行多糖提取.将得到的粗多糖溶液用3倍体积的无水乙醇

目的:通过测定蒙古口蘑多糖(Mongolia Tricholoma polysaccharide,MOP)的抗氧化活性,观察其在低密度脂蛋白(Low density lipoprotein,LDL)被氧化修饰过程中的影响及其在抗内皮细胞凋亡的作用,探讨其可能的保护血管、抗AS的作用机制.方法:将蒙古口蘑子实体用无水乙醇进行脱脂处理后90℃加热6h进行多糖提取.将得到的粗多糖溶液用3倍体积的无水乙醇,4℃过夜沉淀.用Sevage法(氯仿:正丁醇=4∶1)除蛋白,经醇沉淀,依次用乙醇、丙酮和乙醚洗涤.最后得到较纯的多糖.DPPH法检测MOP的清除自由基活性.将LDL分为MOP处理组和未处理组,经Cu2+氧化处理后,ELISA试剂盒检测它们被氧化程度.同时用这两组被氧化的LDL(ox-LDL)刺激血管内皮细胞,用荧光倒置显微镜和流式细胞仪检测细胞凋亡.
