The role and mechanism of histone chaperone Nap1l1 on the regulation of brain development

  Histone variants as important epigenetic regulators play important roles in development.However,it is still unknown that how histone variant H2AZ regulates cortical neurogenesis during brain development.Here we report that the regulatory roles of H2AZ,which is essential for autophagosome formation,in embryonic cortical neurogenesis.We observed that H2AZ expression increased with the brain development and the differentiation proceeding.Suppression of H2AZ expression by shRNAs affected neural progenitor proliferation,differentiation,and cell-cycle exit.The deletion of H2AZ by Cre in H2AZflox/flox mice showed that H2AZ regulated neural progenitor differentiation.Additionally,H2AZ deficiency caused neuronal migration delay and neuronal cell morphology defect.Our findings show that crucial roles of H2AZ in neurogenesis during embryonic cortical development,which may contribute to the explanation of neurodegenerative disorders caused by H2AZ dysregulation.
