The location and characterization of female germline stem cells (FGSCs) in adult swine ovary

  The existence of germline stem cells (GSCs) in adult mammalian female ovaries had been argued since the finding that adult mouse ovaries possessed mitotically active germ cells was proposed.Besides, the location of female germline stem cells (FGSCs) of diverse mammals was at ovarian surface epithelium (OSE) had been reported.Here, we had utilized three methods to detect the location of ovarian GSCs in adult swine, then we cultured these cells and characterized them.First, pluripotent markers (OCT4, NANOG, C-KIT, SOX2, SSEA4 and CD49f) could be detected at the region of theca by immunofluorescence staining, which located outside the basal lamina of granulosa cells.
