  The apical ectoplasmic specialization (ES) and the basal ES are important for germ cell-Sertoli cell communication to support all phases of germ cell development and maturity, and their formation and remodeling require rapid reorganization of the cytoskeleton.However, the molecular mechanism underlying the apical ES and basal ES formation are still largely unknown.Here, we show that the disruption of autophagy in mice Sertoli cells causes male infertile due to the abnormal acrosome biogenesis.In autophagy-deficient Sertoli cells, both β-tubulin and F-actin are disordered, whose classical net structures are disrupted.We find that the degradation of a negative cytoskeleton organization regulator, Pdlim1, is dependent on autuophagy, and it is accumulated in autophagy-deficient Sertoli cells.The disorganization of the cytoskeleton in autophagy-deficient Sertoli cells results in the disruption of apical ES and the basal ES, and finally leads to the malformation of the acrosome.Altogether, this work reveals that the degradation of Pdlim1 by autophagy in Sertoli cells is necessary for maintaining the normal organization of the apical ES and the basal ES, and these findings define a newly recognized role of autophagy in germ cell-Sertoli cell communication.
