PHBHHx膜性材料的表面改性及其与大鼠胚胎神经干细胞的组织相容性研究

  目的 聚羟基脂肪酸酯(PHA)是一类具有良好生物相容性的可降解高分子材料,3-羟基丁酸和3-羟基己酸共聚酯(PHBHHx)是PHA家族中的一员,其性能优于PHA家族的其它材料.研究表明PHBHHx对体外贴壁生长的成骨细胞、软骨细胞、成纤维细胞等多种细胞具有良好的生物相容性,但是对于和体外悬浮生长的神经干细胞(NSC)的相容性欠佳.本研究拟对PHBHHx膜进行表面改性,以增强与NSC的组织相容性,进而为以NSC为基础的脑组织工程应用奠定基础.方法 通过溶剂挥发法制备PHBHHx的二维膜支架,扫描电镜观察其表面性状;分别通过脂肪酶处理,NaOH处理和等离子处理的方法对PHBHHx膜进行表面改性,测量接触角大小检测膜表面亲水性;分离孕14.5天大鼠胚胎大脑皮质NSC,培养于NSC完全培养基(培养基成分:无血清的DMEM/F 12+N2+B27+EGF+bFGF);培养至第三代的NSC分别接种在未处理、脂肪酶处理、碱处理、等离子处理以及碱+脂肪酶处理的PHBHHx支架膜表面,免疫染色观察细胞的分布状态,CCK-8试剂盒检测细胞生长状态,免疫细胞荧光化学观察NSC分化情况.结果 与未处理的PHBHHx膜相比,脂肪酶+NaOH联合处理、等离子处理后,接触角明显减小,提示上述处理可以增强PHBHHx膜表面的亲水性;NSC在接种于PHBHHx支架后,能够继续存活,细胞形态饱满、折光性强,与未做表面改性处理组相比,处理后随着膜亲水性的增强,表面贴附细胞数量增加,存活状态良好,而且能够发生自发分化,分化为神经元和胶质细胞;与PHBHHx支架的共培养促进了NSC的增殖.结论 PHBHHx材料表面亲水性增强可以提高与NSC的组织相容性;与PHHBx材料的共培养,不改变NSC原有的增殖和多向分化潜能.
