干扰骨髓基质细胞Hes1基因对Mash1基因表达情况的影响

  目的 本实验旨在研究干扰体外培养的骨髓基质细胞中Hes1基因后观察Mash1基因的表达变化情况.方法 骨髓基质细胞由全骨髓培养法体外分离培养获得,倒置显微镜下观察其形态变化,应用实时定量PCR技术分析在干扰Hes1基因表达后Mash1基因表达变化情况.结果表明,应用200 nM,100 nM,50 nM,25 nM四种不同浓度梯度的Hes1的siKNA干扰抑制Hes1基因,与对照组相比Hes1表达均降低.结果 200 nM干扰后Hes1基因降低的程度是原来的0.036 (P<0.05),50 nM干扰后Hes1基因降低的程度是原来的0.076 (P<0.05),优于其他浓度的干扰效果.结论 Hes1降低可使Mash1基因表达增加,且Hes1降低越显著,Mashi表达增加越高.所以我们认为,在体外培养BMSCs过程中干扰Hes1基因对Mash1基因的表达有影响,且呈负相关调控关系.
