Objective:This research was to study the hepatotoxicity of Tripterygium Wilfordii Polyglycoside (TWP) on immune inflammation in mice.Method:Mice were respectively given TWP for 5 consecutive days by gavage.The model of ear swelling was made by dinitrofluorobenzene.The effects of drugs on the ear swelling degree and the inhibition ratio were observed.The level of PGE2,TNF-α,IL-2,ALT,AST,PA,TBA,TBIL in serum was detected while the ratio of heart,liver,spleen and kidney to body was measured.Result:The model of ear swelling by croton oil was inhibited by multiple intragastric administration of TWP.The contents of PGE2,TNF-α,IL-2 in serum decreased.The activities of serum ALT,AST,TBA,TBIL and the ratio of liver to body increased.Conclusion:Innmune inflammation can be significantly inhibited by TWP in the dosage range of 6.83~27.3mg·kg-1.The mechanism is related to the generation and decrease of inflammatory medium release.Side effects of different degree can be caused to liver after administration of TWP at the same dosage.All changes of these indexes shows an obvious "dosage-toxicity" relationship.
