Overexpression of Aurora Kinase A (AK-A) has been observed in various tumours and shown to antagonise paclitaxel mediated cancer cell death.Knockdown of AK-A in cancer cells by siRNA synergises with paclitaxel to induce growth inhibition and cell death.We have previously shown that an AK-A specific inhibitor synergises with low concentration of paclitaxel (3nM) to inhibit pancreatic cell growth and cause apoptosis, achieving similar effects to a 10-fold higher concentration ofpaclitaxel (30nM).However, in Human Colony Forming Unit of Granulocyte and Macrophage (CFU-GM) cells, the effect of the combination was simply additive, displaying significantly less myelotoxicity compared to 30nM paclitaxel (60-70% vs 100% inhibition) (Y.Lin, et al.British J.Cancer, 2012).
