轉殖Dehydroascorbate reductase(DHAR)提高單胞藻(Chlamydomonas reinahrdtii)維生素C利用率以增強藻類抗氧化逆境能力

  負責維生素C (Ascorbate,AsA)再生之dehydroascorbate reductase (DHAR)是植物抵抗逆境的重要因子.本研究發現單胞藻(Chlamydomoas reinhardtii)高光(HL,1,800mol·m-2·s-1)處理後3h,DHAR活性和CrDHARl (Cre10.g456750)基因表現量增加,5h後明顯下降,同時伴隨氧化維生素C (Dehydroascorbate,DHA)稹累及AsA氧化還原狀態(AsA/DHA)降低脂質過氧化H2O2過度稹累和細胞死亡的現象,顯示長期高光處理限制DHAR表現而有巖重氧化傷害.因此,我們試圖過度表達CrDHAR1以提高維生素C再生來抵抗高光逆境.E.coli表現系統製作的CrDHAR1重組蛋白,是一種單體(25 kDa),活性79.06mol·min-l·mg-1 protein.透過電穿孔轉殖由HSP70A∶ RBCS2啟動子過度表達CrDHAR1不只增加CrDHAR1蛋白、DHAR活性和AsA/DHA比,也顯示更大的高光抵抗能力及對於methyl viologen (MV)及H2O2誘導氧化逆境之耐受性,高光誘導H2O2產生、 脂質過氧化和光合作用抑制不會發生在CrDHAR1過度表達轉殖株,過度表達CrDHAR1並不會提高維生素C的總量.高光造成之谷胱甘肽(Glutathione,GSH)、氧化谷胱甘肽比例(GSH/GSSG)和Glutathione reductase (GR)和Ascorbate peroxidase (APX)活性之下降,不會發生在QDAR1過度表達轉殖株.以單胞藻系統的結果說明增加AsA再生是藻纇重要抗氧化關鍵.
