The rapid progress of genomics,proteomics and systems biology makes it possible to reverse engineer millions of years of natural evolution and to fashion useful biological systems and products.Such endeavors demand accurate,cost-effective,and high-throughput gene and genome syntheses.Here,I will present our research on new DNA microchip-based technologies for accurate,multiplex gene synthesis and genome assembly and its manifold applications in DNA nanot echnology,protein engineering,molecular evolution,genome-scale circuit rewiring an d perturbation,environmental genomics and genomic medicine.These research efforts will inevitably lead tobetter understanding of the fundamental principles of life and help solve environmental,material,energy,and human health issues.
