目的 :探讨芪龙通络方对2型糖尿病大鼠早期血管病变的保护作用。方法:建立2型糖尿病大鼠动物模型,按随机数字表法分为模型组、二甲双胍组、芪龙通络方组,另取8只大鼠作为正常对照组。各组分别以芪龙通络方水溶液、二甲双胍混悬液、生理盐水灌胃,连续4周后尾静脉采血测血糖,8周后处死动物,股动脉取血,取血清测定血管细胞间黏附因子(sVCAM-1)、血浆测定肿瘤坏死因子(TNF-α)、内皮细胞素-1(ET-1)、一氧化氮(NO)含量。结果:模型组大鼠血清sVCAM-1、血浆TNF-α、ET-1较正常对照组明显升高,血浆NO明显降低。芪龙通络方及二甲双胍均能显著降低sVCAM-1、TNF-α、ET-1及升高NO水平(P<0.05)。结论:芪龙通络方通过减少血管内皮的损伤,从而对糖尿病早期血管病变起到治疗作用,并有助于控制T2DM血管病变的发展。
Objective: To investigate the protective effects of Qi Long Tongluo Prescription on early vascular lesions in type 2 diabetic rats. Methods: Animal models of type 2 diabetic rats were established. They were divided into model group, metformin group and Qilongtongluo Fang group by random number table method. Another 8 rats were used as normal control group. The rats in each group were administered with Qilongtongluo recipe, metformin suspension and saline respectively. Blood was collected from the caudal vein for blood glucose measurement for 4 weeks. After 8 weeks, the animals were sacrificed and the femoral artery was taken for blood. Blood samples were taken for determination of intercellular adhesion molecule (sVCAM-1) and plasma levels of tumor necrosis factor (TNF-α), endothelin-1 (ET-1) and nitric oxide (NO) Results: Serum levels of sVCAM-1, TNF-α and ET-1 in model group were significantly higher than those in normal control group, while plasma NO level was significantly lower. Both QLTT and metformin significantly reduced the levels of sVCAM-1, TNF-α, ET-1 and NO (P <0.05). Conclusion: Qilongtongluo decoction can reduce the damage of vascular endothelium and thus play a therapeutic role in the early stage of diabetic vascular disease and help to control the development of T2DM.
