Multi-drug resistant (MDR) bacteria present a significant,global healthcare problem that is amplified by the lack of new antibiotics,especially against MDR gram-negative pathogens.We developed new pharmacokinetic/ pharmacodynamic models for antibiotics that link binding of antibiotics to their receptors with bacterial responses of multiple bacterial populations (e.g.susceptible and resistant).These models aim at 1) providing a quantitative understanding of the time-course of bacterial killing and mechanisms of synergy of antibiotic combinations against bacteria in specific phenotypic states,2) rationally optimizing antibiotic mono-and combination therapy,and 3) supporting a more efficient design and development of new antibiotics based on receptor binding.
