Proteomics, the analysis of expressed proteins, is becoming rapidly important in the postgenomic era.A primary aim of clinical proteomics is the identification of biomarkers for diagnosis and therapeutic intervention of disease, by comparing the proteomic profiles of disease and control, and differing physiological states.The recent developments of surface-enhanced laser desorption/ionization time-of-flight mass spectrometry (SELDI-TOF-MS) in proteomic research are summarized, and its putative limitations and future applications of disease-specific biomarkers are critically discussed.SELDI-TOF-MS is a novel approach to biomarker discovery that combines two powerful techniques, chromatography and mass spectrometry, with detection sensitivity in the attomole (10E-18) range.This technology is an example of affinity-based laser desorption and ionization spectrometry where proteins of interest are directly applied to a biochemically distinct protein chip array.
