Objective: In the present study, we monitored the antiangiogenesis function of antagomir-21 (miR-21 inhibitor) in tumor angiogenesis in a VEGFR2-luc transgenic mouse model using bioluminescence imaging.Methods: Transgenic VEGFR2-luc mice were used as experiment animals.VEGFR2-luc mice were created using a gene-targeting approach, which "knocked in" firefly luciferasecDNA into the first exon of the endogenous VEGFR2 locus in murine 129/SvEv embryonic stem cells.VEGFR2 expression can drive luciferase gene expression.Murine 4T1 breast cancer cells were purchased from ATCC and were cultured in RPMI1640 medium, with fetal bovine serum at a final concentration of 10%.4T1 cells were implanted beneath the right breast fat pad of VEGFR2-1uc transgenic mice.Tumor-bearing mice were randomly divided into three groups (n=5 in each group), which received saline, antagomir-21, or negative control (scramble)treatment once a week, after tumor model establishment.
