Phosphodiesterases (PDEs) are ubiquitously expressed enzymes involved in regulating intracellular signaling by cleaving the "secondary messengers" cyclic adenosine monophosphate (cAMP) or cyclic guanine monophosphate (cGMP) to their corresponding 5 nucleotides, thus regulating the amplitude and duration of cell signaling mechanisms mediated via cAMP and cGMP.Phosphodiesterases (PDEs) genes constitute a large superfamily, with 21 related genes cloned in mammals to date.Since published works suggest that high intraeellular level of cAMP could arrest growth, induce apoptosis and attenuate cancer cell migration and many cancers pose high levels of cGMP-PDE activity compared to normal tissue, hence use of potent and selective PDE inhibitors should provide novel therapeutic strategies for this enigmatic disease.
