脊髓背角胶状质内结节漏斗部39残基肽对伤害性信息传递的影响

  本研究用盲插全细胞膜片钳技术,在成年大鼠带背根的脊髓薄片上,对结节漏斗部3 9残基肽(tuberoinfundibular peptide of 39 residues,TIP39)在脊髓背角胶状质(substantia gelatinosa,SG)内的作用进行了研究.结果显示:钳制电压为-70 mV,给予TIP39 (0.01-1 μmol/L),在60%的神经元可产生内向电流,且此电流呈剂量依赖性;TIP39引起的内向电流的平衡电位约为0 mV,接近于阳离子的平衡电位;当在电极内液中加入GDP-β-S,全细胞记录形成后30 min左右,此电流可被抑制.另一方面,TIP39对非NMDA受体所介导的微小自发兴奋性突触后电流(miniature spontaneous excitatory postsynaptic currents,mEPSCs)的频率也有增强作用(130.77±12.82%,n=12,P<0.01),而对mEPSCs的幅度没有影响(100.58±5.23%,n=12,P> 0.01)此外,TIP39可增强刺激背根所诱发的兴奋性突触后电流(evoked excitatory postsynaptic currents,eEPSCs)的幅度(140.23±8.35%,n=5,P<0.01),此eEPSCs由A 6和C纤维所介导.以上结果说明TIP39在脊髓背角内对SG神经元有兴奋性作用,此作用由G蛋白偶联受体所介导:同时TIP39也能增强初级传入纤维来的兴奋性突触传递.由此推测,TIP39在脊髓背角SG内增强了对伤害性信息的传递.
