Breast cancer is the most common and the second leading cause of cancer death among US women.We tested whether multiple genetic variations contribute to deficient DNA repair and breast cancer risk using data/samples collected from 752 non-Hispanic Whites (NHW) and 141 African Americans(AA).A total of 18 non-synonymous single nucleotide polymorphisms (nsSNPs) in 4 DNA-repair pathways were tested: (1) Base Excision Repair (BER); (2)Nucleotide Excision Repair (NER); (3) Mismatch Repair (MMR); (4) Double-Strand Break Repair (DSBR).There were significant trends in breast cancer risk with increasing numbers of risk genotypes for ADPRT 762VV, APE1 148DD, ERCC4 415RQ/QQ, and MLH1 219Ⅱ/Ⅳ in NHW and ADPRT 762VA, ERCC2 751KQ/QQ, and NBS1185EQ/QQ in AA, respectively.
