目的:观察阳和汤对兔骨关节炎软骨中基质金属蛋白酶-1(matrix metalloproteinase-l,MMP-1)及基质金属蛋白酶抑制剂-1(tissue inhibitor of metalloprotease-1,TI MP-1)表达的影响。方法:新西兰大白兔20只随机分为正常组、模型组、氨糖美辛组和阳和汤组。按照Hulth法建立膝骨关节炎模型,术后第5周起至第7周末,阳和汤组予混有阳和汤的颗粒饲料,氨糖美辛组给予混有氨糖美辛的兔饲料,正常组及模型组均给予普通饲料;术后第8周分别取各组胫骨平台关节软骨,采用免疫组织化学染色测定软骨中MMP-1及TI MP-1表达的阳性指数。结果:模型组MMP-1及TI MP-1表达的阳性指数水平明显高于正常组,差异有统计学意义(P<0.01),阳和汤组MMP-1及TI MP-1表达阳性指数低于模型组,差异有统计学意义(P<0.05)。阳和汤组和氨糖美辛组MMP-1及TI MP-1表达阳性指数比较,差异无统计学意义(P>0.05)。结论:阳和汤可能是通过调控软骨细胞外基质中MMP-1及TI MP-1表达变化而维持软骨的动态平衡,延缓骨关节炎中软骨退变。
Objective: To observe the expression of matrix metalloproteinase-1 (MMP-1) and tissue inhibitor of metalloprotease-1 (TI MP-1) in rabbit osteoarthritis cartilage with Yanghe decoction. Impact. Methods: Twenty New Zealand white rabbits were randomly divided into normal group, model group, aminoglucose group and Yanghe Decoction group. The knee osteoarthritis model was established according to the Hulth method. From the 5th week to the 7th week after the operation, the Yanghetang group was given a pelleted diet containing Yanghetang, and the ammoniametosyl group was given a rabbit feed mixed with aminomethasine. The normal group and the model group were given normal feed. At the 8th week after operation, the articular cartilage of the tibial plateau of each group was taken. The positive indexes of MMP-1 and TI MP-1 expression in cartilage were determined by immunohistochemical staining. Results: The expression levels of MMP-1 and TI MP-1 in the model group were significantly higher than those in the normal group (P<0.01). The positive expression index of MMP-1 and TI MP-1 in the Yanghe Tang group was low. In the model group, the difference was statistically significant (P<0.05). There was no significant difference in the positive expression index of MMP-1 and TI MP-1 between Yanghe Tang group and Amino sugar group (P>0.05). Conclusion: Yanghe Decoction can maintain the dynamic balance of cartilage by regulating the expression of MMP-1 and TI MP-1 in extracellular matrix of cartilage, and delay cartilage degeneration in osteoarthritis.
