  Restriction endonucleases (REases) and homing endonucleases (HEases) are sequence-specific deoxyribonucleases,REases recognize short DNA sequences with extremely high specificity using multiple contacts per base pair,while HEases recognize very long sites using a few contacts per base pair,thereby allowing for partial degeneracy of the target sequence.They are commonly used as reagents in vitro,and can be used in vivo to introduce,by gene replacement,desired mutations in therapeutically relevant DNA target sites.In the cells,REases and HEases function as selfish genetic elements that are transmitted by horizontal gene transfer.
