A preliminary study of caveolin-3 in the myocardium of type 2 diabetic rats

  Objective To investigate the pathogenesis of the type 2 diabetic cardiomyopathy through preliminarily observing the content of Caveolin-3 (Cln-3) proteins in the injury myocardium of type 2 diabetic rat models.Methods Twenty male SD rats were randomly divided into control group and type 2 diabetic group.Rats were fed with normal diet and high-fat diet respectively.3 weeks later, type 2 diabetic group were provided STZ (25mg/kg)by intraperitoneal injection to induce type 2 diabetes, and normal control group were injected with sodium citrate Citrate buffer solution.After that, rats were continued giving appropriate diets for 4 weeks, then biochemical changes of serum and myocardial tissue were measured.Rats which Fasting plasma glucose (FPG) were above 16.7mmol/L were kept in type 2 diabetic group (n=8).The total protein of Cln-3 was analyzed by Western blot.Results (1) Compared with the normal control group, Lactate Dehydrogenase (LDH) (P<0.01), Creatine Kinase (CK) (P<0.01), Maleic Dialdehyde (MDA) (P<0.05) and Fasting Serum Insulin (FINS)(P<0.01) were significantly increased, while Superoxide Dismutase (SOD) (P<0.01) and Insulin Sensitive Index (ISI) (P<0.01) were significantly decreased in type 2 diabetic group.(2) Western blot showed that, the Cln-3 proteins were significantly increased in the injury myocardiums of type 2 diabetic group (P<0.05).Conclusion Cln-3 protein is significantly increase in the injury myocardium of type 2 diabetic rat.
