  Highly sensitive separations of biogenic compounds by capillary electrophoresis (CE) and microchip electrophoresis (MCE) using an on-line sample concentration technique, i.e., large-volume sample stacking with an electroosmotic flow (EOF) pump or LVSEP [1,2], were investigated.In the LVSEP-MCE analysis of oligosaccharides released from bovine ribonuclease B, more than 2000-fold improvement of the detectability was achieved compared to a conventional MCE analysis using the pinched-injection.In LVSEP-CE of oligosaccharides, ca.780-fold sensitivity enhancement was attained without any loss of resolution compared to a conventional CE analysis.To make further sensitivity improvement possible, LVSEP was combined with the field-amplified sample injection (FASI) technique.Maltoheptaose (G7) derivatized with 9-aminopyrene-1,4,6-trisulfonic acid (APTS) was analyzed by LVSEP-FASI-CE.As a typical result, G7 was well concentrated and separated from the free APTS with a 10,000-fold sensitivity increase compared to the conventional CE analysis, where the good repeatability of the migration time (%RSD =2.4; n =3) was obtained.
