A review on the conditional miRNA biogenesis enzyme knockout mice

  Objective The non-coding miRNAs are processed in a step-wise manner from the genomic DNA sequences through pri-miRNAs and pre-miRNAs to the matured 21-25bp nucleotides(usually 22bp),during which course three enzymes referred to as Pasha,Drosha and Dicer(Pasha and Drosha exist in the nucleus while Dicer in the cytoplasm)play the key roles for the miRNA biogenesis.Therefore,the deletion of each of the above enzymes will block the miRNA biogenesis pathway,causing the down expression of most miRNAs and resulting in the animal embryonic defect.However,it is surprisingly found that the knocked out animals of Pasha,Drosha and Dicer have different phenotypes from one another,indicating the three enzymes may have their respectively additional roles in animals.Till now several tissue-specific conditional miRNA biogenesis enzyme knockout mice have been established to determine the roles of the enzymatic genes in different tissues,which further expounds the related miRNA functions in whole animal bodies.
