Ninety percent of all ophthalmic tumors arise in periocular skin, and if left untreated, these ocular surface lesions can progress to life-threatening metastasis.Although surgery excision remains the most effective therapy for these tumors, development of non-surgical treatment is necessary for patients who cannot tolerate surgery or whose vital periorbital structures need to be preserved.Previous studies have demonstrated that gain-of-function mutations in FGF signaling can drive benign skin tumor in mice and humans, but interestingly, genetic evidence also shows that deletion of FGF receptor causes epidermal thickening and spontaneous papillomas, suggesting a protective role of FGF signaling in skin tumor.In this presentation, I will discuss our recent findings in a mouse model of squamous cell carcinomas in periocular skin, and show that FGF signaling is induced in an autocrine fashion to promote cell proliferation and tumorigenesis.Importantly, inhibition of FGF signaling completely suppresses skin tumor formation in our model.Our results thus suggest that components of FGF signaling may be therapeutic targets for treating periocular skin tumor.
