Notch signaling plays a critical role for specifying the intestinalepithelial cell lineage and for initiating colorectal adenomas andcolorectal cancers (CRC).JAG2 as a ligand of Notch signaling pathway,is involved in breast cancer metastasis and multiple myelomadevelopment.But its role in colorectal cancer remains still unclear.In this study,we examined the expression patten of JAG2 in humannormal colonic cells and CRC cell lines.We found that JAG2 wasoverexpressed in all CRC cell lines tested.Furthermore,silencingJAG2 by using short interference RNA (siRNA) inhibits cell invasion butnot affects cell proliferation in CRC cell lines.But inactivation ofNOTCH signaling pathway didn’t decrease cell invasion by using notchpathway inhibitor N-[N-(3,5-difluorophenacetyl)-1-alanyl]-S-phenylglycine t-butyl ester (DAPT),suggesting inhibition of NOTCH signaling is insufficient to attenuate invasion of CRC cells.Notably,suppression of cathepsin K,a cysteine protease of the papain family that cleavescollagen,may contribute to JAG2 depletion mediated downregulation of cell invasion.
