大蒜素对IL-6诱导的胃癌细胞EMT的作用研究

  目的 通过建立胃癌细胞EMT 模型,探讨大蒜素对IL-6 诱导的胃癌细胞上皮-间质转化(EMT)的抑制作用.方法 常规培养胃癌细胞株HGC-27 细胞,取对数生长期细胞,分别以50ng/mL 白介素-6(IL-6),50ng/mL IL-6+3μ g/L 大蒜素,3μ g/L 大蒜素处理胃癌HGC-27 细胞,另设空白对照组,通过Cell Counting Kit-8(CCK-8)法检测细胞增殖情况;划痕试验、Transwell 实验测定胃癌细胞的迁移、侵袭的作用.结果 CCK-8 结果 显示,与空白对照组相比,大蒜素组对胃癌HGC-27 细胞的增殖具有明显的抑制作用(P<0.001),IL-6 干预组具有明显的增殖优势.划痕试验结果 显示:IL-6 组、空白对照组、大蒜素组、大蒜素联合IL-6 组12h 划痕愈合百分比(%)分别为23.50±3.49、27.78±6.72、5.70±3.85、14.53±7.16;24h 划痕愈合百分比(%)分别为51.06±2.11、46.60±2.10、29.30±3.07、36.43±0.96.在给药前的0h 时,各组细胞的伤痕宽度无显著性差异(P>0.05).随着时间推移,划痕开始得到修复,与空白对照组比较,大蒜素组胃癌HGC-27 细胞划痕愈合百分比明显下降;与IL-6 组相比,大蒜素联合IL-6 组胃癌细胞的划痕愈合百分比明显下降(P 均<0.001).Transwell 实验结果 显示,与空白对照组相比,大蒜素组侵袭数目明显减少,且大蒜素联合IL-6 组细胞的侵袭能力较对IL-6 组明显减弱,差异有统计学意义(P 均<0.001).结论 大蒜素可能抑制IL-6 诱导的胃癌细胞EMT 的作用,进而在肿瘤浸润转移中发挥重要作用.
