Argonaute蛋白家族可分为AGO和PIWI两个亚家族成员,它们在物种间高度保守,并在小分子非编码RNA调控途径中发挥核心作用.其中AGO蛋白家族成员特异性地与small interfering RNAs (siRNA)和microRNAs (miRNAs)结合,在多种组织器官中发挥重要功能.而piwi(P-element-induced wimpy testis)最初是在果蝇中通过P因子增强子捕获技术筛选影响生殖干细胞分裂的基因时发现的.研究人员通过在果蝇,小鼠,线虫,斑马鱼等模式生物中将piwi基因进行功能缺失突变,发现PIWI蛋白在动物生殖细胞发育和配子生成过程中发挥多种重要功能.然而PIWI蛋白并不是单独来发挥这些功能的.2006年,四家独立的研究小组几乎同时在果蝇、小鼠、大鼠和人等物种生殖系细胞中发现了一类新的小分子非编码RNA.研究人员通过特异性的PIWI蛋白抗体对睾丸组织的全细胞裂解液进行免疫共沉淀实验,他们发现PIWI蛋白可以特异性地结合一类24-33nt的小分子RNA,因此这些小分子RNA被命名为PIWI相互作用RNA(PIWI-interacting RNA),简称piRNA.
