Analytical approaches with multi-resolution are highly desirable for extracting more information of the components from a sample under analysis.Herein, a new concept, spatio-temporally resolved capillary electrophoresis (STRCE), is presented for the first time.Analytes are separated within the separation capillary with spatial resolution as in conventional CE, whereas the whole capillary is detected with a dynamic detector such as a CCD camera, which offers a temporal resolution for the analytes.STRCE can offer several significant advantages over conventional CE and is particularly useful for analyses with kinetic measurements such as studies on biomolecular interactions. The concept and applications of STRCE are demonstrated with several recent research works on capillary isoelectdc focusing (CIEF) whole column imaging detection (WCID) by the authors.Important applications include two-dimensional protein characterization and protein-DNA binding study. The former allows for fast determination of molecular weight and isoelectric point for proteins while the latter permits the discrimination of different interaction types and the measurement of the individual dissociation rate constants.Future development of this approach is also highlighted.
