We investigated the effect of Tetramethylpyrazine (TMP), one of the main active compounds isolated from Chuanxiong which has significantly neuro-protective effect, on micro-vascular endothelial cells (BMECs) damage induced by oxygen-glucose-deprivation (OGD).Recent studies have shown that Rho-kinases play a key role in the pathogenesis of ischemic stroke.As an extending study we investigated whether the neuro-protective effect of TMP is mediated by mitigating the active of Rho-kinases thereby reducing oxidative stress.It was found that the presence of TMP decreased Reactive Oxygen Species (ROS),one vital product of oxidative stress which can lead to BMECs contraction and increase its permeability, by down-regulate the active Rho-kinases in OGD-induced BMECs.Furthermore TMP maintain the integrity of cytoskeleton in OGD-induced BMECs by down-regulate RhoA-kinase protecting the integrality of blood-brain barrier (BBB).The results suggest that TMP exert neuro-protective effect in OGD-induced BMECs via down-regulating the active of Rho-kinases, reducing oxidative stress and protecting the integrality of BBB.
