A new thermophilic inulinase producing strain,which grows optimally at 60℃,was isolated from soil samples on medium containing inulin as a sole carbon source.It was identified as a Bacillus smithii by analysis of 16s rDNA.Various parameters of fermentation,such as ingredients of medium,processing parameters have been examined in order to improve enzyme yield.Inulinase yield of 135.2 IU/ml was achieved with medium pH7.0,containing 2.0% inulin,0.5% (NH4)H2PO4,0.5% yeast extract,0.5mM MgCl2,0.006% Brij-35 at 50℃ 200rpm shaker,6% inoculation volume and 72h incubation.The enzyme shows endo-inulinolytic activity,high activity at pH 4.5 and is stable among pH range of 4.0-8.0,whereas the optimum temperature for enzyme activity was 70℃,49% and 33% activity remained after 9 h at 70℃ and 80℃ incubation respectively.This finding is significant for its potential industrial application.
