目的:miR-291 b-3p是miR-290家族中的一员,在胚胎发育过程中起重要作用,而在脂代谢中的作用尚未见报道,本研究旨在阐明其在肝脏脂质代谢中的作用及其机制.方法:首先在NCTC 1469细胞内通过油酸/棕榈酸(2∶1)刺激24h造成细胞内脂质蓄积,油红O检测细胞内脂质蓄积情况,western blot检测脂代谢相关蛋白的表达,q-PCR检测miR-291b-3p在细胞内的表达.其次在细胞脂质蓄积模型内转染miR-291b-3p的inhibitor,油红O检测细胞内脂质蓄积情况,western blot检测脂代谢相关蛋白的表达.再次通过软件预测寻找miR-291b-3p潜在的靶基因,通过荧光素酶活力分析及western blot检测其与靶基因的关系.
