AEZS-108 (AN-152) is a cytotoxic analog of the luteinizing hormone releasing hormone (LHRH) in which doxorubicin (DOX) is linked to the LHRH agonist D-Lys(6).AEZS-108 binds to LHRH-receptors, which are found on a variety of tumors including breast, prostate, ovarian and endometrial cancers.After binding, AEZS-108 is internalized and transported to the nucleus where it induces apoptosis upon release of DOX.The activity of this compound has been demonstrated in experimental xenograft models of a variety of human cancers.Targeted chemotherapy with AEZS108 was significantly more effective and less toxic than DOX itself without induction of the MDR1 protein.A phase Ⅰ study demonstrated partial remission and stable disease in patients with LHRH positive gynecological cancer progressing following conventional therapy.Results of phase Ⅱ studies in ovarian and endometrial cancer confirmed good clinical response with remarkable overall survival and a favorable side effect profile.
