Objective: To investigate indoleamine 2,3-dioxygenase (IDO)expression in myeloid-derived suppressor ceils (MDSCs) in breast cancer and its function in MDSC-mediated inhibition of immune surveillance.Methods: Clinical samples were collected from 85 primary breast cancer patients who received radical mastectomy at the Department of Breast Oncology of Tianjin Medical University Cancer Institute and Hospital, from January 2009to December 2009.CD33+ cells were isolated from breast tissues and peripheral blood mononuclear cells (PBMCs) usingCD33 MicroBeads; in addition, flow cytometry examined the immunophenotype and further sorted these cells.CD3+ T cells were prepared from 10 cases of healthy PBMCs using human pan T cells isolation kit Ⅱ.Two nonspecific T cell polyclonal amplification protocols and stimulation using IL-2 and antiCD3/CD28 mAb were used to evaluate the immunosuppressive activity of fMDSCs and iMDSCs on T-cell proliferation,cytokine secretion, and apoptosis.Immunohistochemistry(IHC) staining of 50 cases of formaldehyde-fixed, paraffinembedded breast tissues was performed as previously reported.
