Background: Epithelial cell adhesion molecule (EpCAM) is a type I transmembrane glycoprotein with molecular weight of 40 kDa.It is highly expressed in human epithelioma, but lowly expressed in normal epithelial tissues.Objective: To construct an EpCAM-specific recombinant immunotoxin and further test its antitumor activity.Methods: The immunotoxin (GST-scFv2A9-PE or GST-APE) was constructed by genetically fusing a truncated form (PE38KDEL) of Pseudomonas aeruginosa exotoxin with an anti-EpCAM single chain fragment variable (scFv2A9).
