Abscisic acid plays a major role in plant growth and development, and in adaptation to environmental stress.The research on abscisic acid signal transduction is remarkable and has advanced rapidly in recent years.A major mechanism of signal transduction is reversible protein phosphorylation mediated by protein kinases and protein phosphatases involved in regulating many biological processes.The PP2C-type protein phosphatases (PP2C) are a kind of Ser/Thr Protein phosphatases (PPs), participating extensively in ABA signal transduction.PP2C were found to be the regulators in the pathway.PP2C gene homologs have been isolated from many species, including Arabidopsis, Maize and Beech.A full length cDNA of PP2C gene from Moso bamboo (phyllostachys edulis) was cloned by RT-PCR.It had 990 bp nucleotides and coded 330 amino acid residues.To study its functions, the coding region of PhePP2C was introduced into a vector with the CaMV35S promoter and then transformed into wild-type Arabidopsis thaliana.The expression of PP2C in transgenic plants will be tested by QRT-PCR to analyze its functions.ABA content in different organs from Moso bamboo will be detected using High Performance Liquid Chromatography (HPLC) to analyze the function of PhePP2C gene in ABA signal transduction pathway.
