  Background: Human epidermal growth factor receptor type 2 (HER2) overexpression is associated with accelerated disease progression and poor prognosis in breast cancer (1,2,3).Several studies have found significant influence on the results for the determination of HER2 expression with different analytical methods i.e.Immunohistochemical staining (IHC), fluorescence in situ hybridization (FISH) or polymerase-chain reaction analysis (PCR) (4,5,6,7,8).In a prospective study, which has not previously been reported, we compared the influence of two different tissue preparation techniques, laser assisted-microdissection (LMD) and whole tissue paraffin macrodissection (MAC), on her2 gene relative quantification (her2RQ) results.Methods: The influence of two different tissue techniques, i.e.LMD (ArcturusXT LCM system) or MAC, on her2RQ (LC-HER2/neu DNA Quant.Kit, Roche) (9) results, were investigated in formalin-fixed paraffin embedded tissue from 97 women with breast cancer.A relative increase in the number of gene copies of two or more was considered to indicate HER2 amplification.We also compared LMD followed by her2RQ with IHC (DAKO HercepTes) (10).Results: A significant correlation was found between the quantitative HER2 gene results with the two different tissue preparations: LMD or MAC (R =0.64 P< 0.001).
