场强放大堆积-毛细管电泳法测定丹参不同部位的五种水溶性有效成分

利用高效毛细管电泳-场强放大柱内堆积技术分离测定白花丹参中的水溶性有效成分原儿茶醛、原儿茶酸、丹参素、迷迭香酸和丹酚酸B采用未涂层熔融石英毛细管柱(50.2 cm×75 μm,有效长度40 cm),以15 mmol/L硼砂-15%甲醇(pH=10.0)为运行缓冲液,分离电压28 kV,柱温25℃,检测波长210 nm进行测定.进样前压力进水0.5 psi×4 s;电动进样-8 kV×3 s.原儿茶醛、原儿茶酸、丹参素、迷迭香酸和丹酚酸B的线性范围分别是3.0 μg/mL~60.0 μg/mL(R2=0.9996),1.0 μg/mL~20.0 μg/mL(R2分别为0.9991、0.9994、0.9989和0.9998),检测限分别为0.55 ng/mL,0.40 ng/mL,0.25 ng/mL,0.32 ng/mL和0.38 ng/mL,富集倍数高,精密度良好;将此方法应用于丹参不同部位五种水溶性有效成分测定,回收率为97.31 %~99.87 %.该方法简单、快速、准确、灵敏度高,可用于丹参的质量控制;而且丹参水溶性成分的标志物丹酚酸B的含量呈根>叶>花>茎的趋势,其叶、花和茎都有较高的药用价值.
