Cysteinyl leukotrienes (CysLTs) are 5-1ipoxygenase (5-LOX) metabolites of arachidonic acid and exert potent pro-inflammatary effects via their receptors including cysteinyl leukotriene receptors 1 and 2.We recently found that the expressions of 5-LOX,cysteinyl leukotriene receptors 1 and 2 in the reactive astrocytes were increased in rat brain after focal cerebral ischemia.However,in the cultured astrocyte,whether 5-LOX/CysLTs/CysLT1 receptor pathways are also involved in glial response induced by ischemic injury,whether CysLT2 receptor also participates in astrocyte response remains unknown.In the present study,we used 1-h oxygen-glucose deprivation as sub-lethal injury to induce the reactive gliosis; and 4-h OGD as moderate insult to induce cell injury in primary cultured astrocytes.We found 5-LOX was activated and participated in both the reactive gliosis and cell injury of astrocytes induced by OGD and recovery.
