This paper provides scientific basis for the evaluation of the antidepressant efficacy and the mechanism of action through liquid chromatography-mass spectrometry(LC-MS)technique to analyze the plasma samples of patients with depression before and after Xiaoyaosan and healthy control group.LC-MS-based metabolomics techniques and multivariate statistical methods were used to analyze the plasma metabolites to find potential biomarkers related to antidepressant efficacy of Xiaoyaosan.The plasma levels in depressed patients change significantly compared with healthy controls,a total of 10 potential biomarkers related to antidepressant efficacy of Xiaoyaosan were found,and the content of the most of these endogenous metabolites were returned to normal levels after the treatment of Xiaoyaosan.Application of these biomarkers in clinical practice may help to optimize the diagnosis of depression and to evaluate the efficacy of Xiaoyaosan.Metabolomics will be a powerful tool for the discovery of biomarkers.
